Florida Hospital Pepin Heart Institute and University of South Florida team up for the gene therapy trial
TAMPA, Florida (Feb. 21, 2013) Cardiovascular disease specialists at Florida Hospital Pepin Heart Institute and Dr. Kiran C. Patel Research Institute affiliated with the University of South Florida announced they have enrolled their first patients into a clinical trial testing a novel gene therapy for the treatment of heart failure after ischemic injury. The therapy may promote the regeneration of heart tissue by encouraging the body to deploy more stem cells to the injury site.
Dr. Charles Lambert, Medical Director of Florida Hospital Pepin Heart Institute and Dr. Leslie Miller, Director of the USF Heart Institute, are leading the way for the randomized, placebo-controlled trial which spans 10 sites across the United States. The study, called the STOP-HF, will enroll 90 patients nationwide.
Heart failure (HF) can occur when the muscles of the heart become weakened and cannot pump blood sufficiently throughout the body. The injury is most often caused by inadequate blood flow to the heart resulting from chronic or acute cardiovascular disease, including heart attacks. Considerable scientific evidence has emerged over the past decade demonstrating the high therapeutic potential of regenerative medicine for a host of diseases. Heart failure is a leading cause of death, disability and hospitalization.
Dr. Charles Lambert is performing the gene therapy by direct injection into the heart using an investigational system in the catheterization laboratories at Florida Hospital Pepin Heart Institute.
"Pepin Heart and Dr. Kiran C. Patel Research Institute and USF are exploring and conducting leading-edge research to develop break-through treatments long before they are even available in other facilities," Dr. Lambert said. "Stem cells have the unique ability to develop into many different cell types, and in many tissues serve as an internal repair system, dividing essentially without limit to replenish other cells. This trial is unique in that it uses gene therapy to turn on a process leading to cell regeneration rather than simply administering stem cells directly."
The Pepin Heart Institute has a history of cardiovascular stem cell research as part of the NIH sponsored Cardiac Cell Therapy Research Network (CCTRN) as well as other active cell therapy trials. Locally, the STOP-HF trial is the first of several regenerative medicine clinical trials teaming the USF Heart Institute with Florida Hospital Pepin Heart Institute, which is adjacent to the USF Health campus.
"This is the beginning of a new era in cardiovascular therapies," said Dr. Leslie Miller, national principal investigator for the STOP-HF trial and professor of cardiovascualar sciences at the USF Health Morsani College of Medicine. "Targeted gene and cell therapies delivered directly into the heart hold promise for helping to regenerate tissue, reduce injury and restore heart function. USF Health, working with our partners, will find new ways to diagnose and treat patients, with the aim of reducing and ultimately harnessing the global impact of heart disease."
The trial, sponsored by Juventas Therapeutics, is a double blind, randomized, placebo-controlled Phase II study evaluating the safety and effectiveness of JVS-100 in patients with ischemic heart failure. JVS-100 is the name of the gene therapy that directs the heart muscle to produce Stromal cell-Derived Factor 1 (SDF-1), a protein that has been shown to repair damaged tissue in the body through the recruitment of circulating stem cells to the site of injury, prevention of ongoing cell death and restoration of blood flow.
Earlier this year, Juventas reported results from its Phase I study in Class III ischemic heart failure patients. In addition to meeting the primary safety endpoint, patients in the study who received the drug demonstrated clinically significant improvements in exercise levels at the 12 month mark.
Other prominent institutions participating in the multicenter trial include Columbia University Medical Center, The University of Utah, the Lindner Center for Research at the Christ Hospital, and the Minneapolis Heart Institute Foundation.

(Reuters) - The Academy Awards are usually tightly scripted events, but sometimes even Oscar gets a taste of the unexpected. Here are some of the curious moments when things veered off course on the red carpet.
REFUSING OSCAR
A few Oscar winners have felt compelled to turn down their prizes over the years. The first was screenwriter Dudley Nichols, who refused his Best Screenwriter award for "The Informer" in 1936 because of conflicts between the Screen Writers Guild and the Academy. Marlon Brando famously sent a proxy to refuse his 1972 Best Actor Oscar for "The Godfather" on his behalf (and to deliver a 15-page speech on Hollywood's mistreatment of Native Americans while she was there).
The all-time greatest rejection, though, goes to George C. Scott, who denounced his Best Actor nomination for "Patton," calling the awards "offensive, barbarous and innately corrupt." Scott was quoted as calling the ceremonies "a two-hour meat parade, a public display with contrived suspense for economic reasons." When he won the award, he was 3,000 miles away at home, watching a hockey game on TV. Brilliantly, Scott's high moral tone was strangely absent when, two years later, he let it be known that if the Academy felt like nominating him for Best Director for "Rage," he wouldn't object. Unsurprisingly, they didn't much feel like it.
Ten years later, Scott was so keen on the idea of an offensively barbarous two-hour meat parade that he bought last-minute tickets for the 1982 ceremony, and would have gotten away without incident had he not been spotted by a columnist from Variety and heckled on the red carpet.
A LONG WALK TO THE STAGE
Hattie McDaniel was the first African-American to win an Academy Award, for her portrayal of Mammy in the 1939 film "Gone With The Wind" - as mentioned by George Clooney in his 2006 "Isn't Hollywood progressive?" Oscars speech. (https://www.youtube.com/watch?v=NqDbG9h-f7c ) What he didn't mention was that, while the rest of the cast and crew of "Gone with the Wind" sat at a big table together, McDaniel and her companion were seated at a table for two in the back of the room, as the Ambassador Hotel was still segregated. And it was from all the way at the back of the room that she had to walk to accept her award with, one has to say, a very gracious speech under the circumstances. MINISKIRT BAN
In her 1967 classic "How to Dress for Success," Oscar-winning designer Edith Head said, "Even the most beautiful legs - Marlene Dietrich's, for instance - look better when the kneecap is covered." To back up the sentiment, Head, who served as a special adviser to the Academy, banned miniskirts from the 40th Academy Awards in 1967, saving the nation from the indignity of knees it suffered in 1966 (like the shocking mid-leg areas of Inger Stevens: http://www.ingerstevens.org/images2.html - although to be fair, that IS more of a swimsuit than evening wear).
THE STREAKER
Daring to show a little more leg (and a lot more of everything else) than even the poor, oppressed miniskirt lovers of 1967 was Robert Opel, who in 1974 became the only person to have appeared naked onstage at the Oscars. (http://www.youtube.com/watch?v=2IIl3zSYL8k ) Sadly, he wasn't a daring nominee, or even a presenter - just some guy who really liked being naked. "Isn't it fascinating to think that probably the only laugh that man will ever get in his life is by stripping off his clothes and showing his shortcomings," quipped David Niven, securing his place in dictionaries of hilarious quips forevermore. Of course, it was later suggested that the streak had been arranged by the producers to spice up a boring run of ceremonies, and that Niven has been seen to borrow a pencil to note down his spontaneous line before the show had even begun, but it's far more fun to believe the opposite.
THERE'S NO HOSTING LIKE ... WELL, NO HOSTING
There have been good hosts, bad hosts, really bad hosts and really, really bad hosts, but the all-time worst ceremony was one with no host. Few people who saw the 1989 awards show's opening number - a duet between an actress playing Snow White and non-singer Rob Lowe - will ever forget it. ... no matter how much they might want to, or how hard they might try. It wasn't the first time the show had gone without a host; there was a three-year run in the late '60s after Bob Hope's tenure, when no one could follow his masterful act. But 1989 was the last time that happened.
BANNED!
After the Academy caught wind of Sasha Baron Cohen's plan to turn up dressed as his leading role in "The Dictator," rumors flew in the weeks leading up to the 2012 Oscars that Baron Cohen would be barred from the ceremony. The Academy, however, said he had never been banned, just warned that the red carpet was no place for stunts. Apparently, Baron Cohen didn't get the message; he arrived in full costume, carrying an urn containing, he said, the ashes of Kim Jong-il, which he then proceeded to spill over Ryan Seacrest before being escorted off the premises. Actual bans from the Oscars are harder to come by - but not impossible. Just ask Nicholas Chartier, who was reportedly the first nominee ever to fall that far afoul of Oscar, for sending an email to a group of people including Academy members encouraging them to support the film he produced ("The Hurt Locker") and disparaging another nominated film. VALUE OF THE WORLD'S MOST FAMOUS STATUETTE: $1
Much to the disappointment of some faded stars, there's no profit to be made from an old Oscar. Since 1950, the Academy has made every recipient of the little golden man sign a "winner's agreement." If you fall out of love with fame and one day wish to sell you statuette, you have to offer it to the Academy first for $1. That doesn't mean that Oscars never surface in auctions - Steven Spielberg has spent more than $1.1 million on two used pre-winner's-agreement Oscars - Clark Gable's in 1996 and Bette Davis' in 2001, in order to return them to the Academy. There's nothing someone with a shelf of real Golden Men likes less than someone else trying to buy their way into the club.
THE SHIFTING STATUETTE
The modern award is 13.5 inches high, weighs 8.5 pounds (3.85 kilograms) and is made of gold-plated britannium, a pewter-like alloy, on a black metal base. But it has not always been so. Before World War Two, the base was stone; during the war, statuettes were made of plaster as a nod to the war effort (though winners could swap them for metal ones once the war was over).
Until the 1950s, child actors who won Oscars were given miniature statuettes. This was not because they were too tiny and weak to carry the big ones off the stage but because it was thought unfair to the adults that they would have to compete with kids.
When ventriloquist Edgar Bergen and his dummy Charlie McCarthy got an honorary Oscar in 1938, he was given a wooden Oscar statuette with a movable mouth.
AUSTERITY OSCARS
In 1944, as part of the war effort, the Academy took the 16th Academy Awards to Grauman's Chinese Theater, the first time for the Oscars in a big public venue. Men and women in uniform were given free tickets for the ceremony, and in a show of solidarity some were given seats on the stage. THE OSCARS' OSCAR
The late Walt Disney currently holds the record for winning the most, with 26 awards given to him personally (22 Oscars, three Special Awards and the Irving G. Thalberg Memorial Award).
On the flip side, sound re-recording mixer - it's a real job, apparently - Kevin O'Connell has been named the Oscar's most unlucky nominee, having been nominated 20 times without winning.
Meanwhile, there has only ever been one Oscar to win an Oscar. Songwriter Oscar Hammerstein II, fittingly, won two.

Country singer Mindy McCready was found dead Sunday "from what appears to be a single self-inflicted gunshot wound," police said.
Deputies investigating a report of gunshots found McCready's body on the front porch of a home?in Heber Springs, Ark., according to the?Cleburne County Sheriff's Office.
McCready, 37, had battled substance abuse.?
?As sad as it is, it didn?t come as a major shock, because she?s just been battling demons for so long," Billy McKnight, McCready?s ex-boyfriend and the father of her 6-year-old son, told TODAY on Monday. ?I was around her when she attempted suicide twice, so I knew it was in her.?

She had a No.1 country hit in 1996, when she was just 21, with "Guys Do It All the Time." Her other popular songs included "Ten Thousand Angels" and "A Girl's Gotta Do (What A Girl's Gotta Do)."
"I grew up listening to?Mindy McCready...so sad for her family tonight," Grammy-winning country singer Carrie Underwood tweeted. "Many prayers are going out to them."
Wynonna Judd tweeted, "Oh my! Mindy. Dear sweet girl. This is so sad. It just breaks my heart what addiction continues to take from this life."
McCready, who has dated such famous men as "Lois and Clark" star Dean Cain and Red Sox pitcher Roger Clemens, has had more problems than hits in recent years.

Her relationship with Clemens made headlines in 2008. McCready told "Inside Edition" that she was just 16 when she met the married Clemens, but said their relationship did not become sexual until years later. Clemens issued a statement saying only, "I have made mistakes in my personal life for which I am sorry."
On Monday, Clemens released a statement calling McCready's death "sad news," and adding, "I had heard over time that she was trying to get peace and direction in her life."
In 2010, she appeared on "Celebrity Rehab with Dr. Drew" seeking help for her battle with substance addiction.
Her biography for the VH1 show states, "Although she was arrested and jailed for trying to buy OxyContin with a false prescription and attempted suicide with an overdose of drugs and alcohol, Mindy?believes her only true addiction is to violent relationships."
In an interview with The Associated Press in 2010, McCready called her life "a beautiful mess."?
She added: "My entire life things have been attracted to me and vice versa that turn into chaotic nightmares or I create the chaos myself."?
In 2011, McCready's mother reported that the singer and her young son Zander were missing. McCready's mother was Zander's legal guardian. The singer returned her son by order of a judge. She gave birth to another son, Zayne, in April 2012.
In January, her boyfriend David Wilson died of a gunshot wound in his Heber Springs home.?McCready later appeared on TODAY and called Wilson her "soulmate." When asked by NBC's Andrea Canning if she shot Wilson, McCready replied, "Oh, my God, no. He was my life. We were each others' life."
Authorities are still investigating Wilson's death and have not named McCready as a suspect.
On Feb. 6, McCready's publicist said the singer was admitted to an inpatient facility, though the cause was not specified. Her two sons were placed in foster care while McCready was being treated.

Sounds like a stretch, but the Goog is breaking out of its search box big time, and recent additions to the Nexus line are proving it has a real chance at establishing a direct relationship with customers.
Let us initially take a small peek into the very foundation of this world renowned city and as to when it was built.
As one reflects on the glorious past it is evident that the first of the settlers happened to approach Tikal in South America during the 10th century BC.

While one gets over with the reaction of the grandeur of Temple I the Masks Temple the Temple II stands proudly at 122 feet tall facing the east, known to be used by Jasaw Chan Kawiil, during his lifetime.
Housing the remains of the altars that leave tell-tale signs of the acoustically designed temple, it enabled the hearing of the high priests then, which is today demonstrated by the guides who clap their hands to demonstrate the same.
The echo of the clapping hands leave the listeners all inspired and starry eyed at the magnificence of the designers and the intellect used behind the designing and its multifaceted purposes.
The story goes on though mingled with facts and beliefs, after all certain realities get buried with the ones who held the reins!
